Of Isleham, Sheriff of Cambridgeshire and Huntingdonshire, Knight of the Sire for Cambridgeshire.
Son and heir to Sir Robert Peyton and Frances Hasilden. Grandson of Sir Robert Peyton of Wicken by Elizabeth Clere.
Husband of Elizabeth Rich, seventh daughter of Richard Rich, Lord Rich of Leighs Priory Essex and Elizabeth Jenks, daughter of William, a London grocer. They were married before 1557 and had three sons and three daughters; Robert, Sir John, Richard, Mary, Frances and Winifred.
Robert was knighted at Royston in Nov of 1608 by King James I.
Robert and Elizabeth were buried in the South Chapel of Isleham with an alabaster tomb with effigies and monumental inscriptions.
